Antioxidant and Genoprotective Activity of Rutin and Quercetin from Citrus Fruits

Free radicals impair the cellular and tissue homeostasis, which can cause several clinical manifestations such as cancer, Alzheimer, cardiovascular diseases and so on. Rutin and quercetin are two important phenolic compounds found in a variety of sources such as citrus fruits, propolis, grapes, tomatoes, berries etc. These phenolic compounds probably are effective in treatment of various disorders resulted from free radical mediated injuries. In the present study, free radical activity of these compounds by DPPH and H2O2, cytoprotective activity by MTT assay and Geno protective assay by COMET assay. The cytotoxic effects of the compounds were evaluated by MTT assay on rat’s lymphocytes. COMET assay was performed for DNA damage analysis. Cytotoxicity and genotoxicity were induced by well-known cytotoxic chemical acrylamide, formed during the processing of high temperature prepared food. The results of the present study indicate that both compounds exhibited a good percentage of free radical scavenging activity against radicals of DPPH and H2O2. The cytoprotective activity of these compounds showed a good effect evident by cell viability percent calculation. Geno protection was also evident by calculating tail damage, tail moment and % DNA. Therefore, rutin and quercetin probably is a favourable option in drug discovery against the free radical mediated clinical manifestations and their resulted consequences.
